چکیده

Introduction Parkinson’s disease is a progressive neurodegenerative movement disorder characterized by reduction and slowness of movement (akinesia and bradykinesia), muscle rigidity, and tremor. The core pathology of the disease is the degeneration of midbrain dopaminergic neurons in the substantia nigra pars compacta that project to the striatum and other basal ganglia nuclei. The striatum is the main input structure of the basal ganglia and receives excitatory projections from many cortical areas and thalamic nuclei. The output neurons of the striatum, named as medium spiny neurons, are g-aminobutyric acidergic (GABAergic) and project to both the globus pallidus internus and externus, and to substantia nigra pars reticulata, which are also GABAergic. The other nucleus within the basal ganglia is the subthalamic nucleus that receives direct cortical inputs and an input from the globus pallidus externus and its glutamatergic neurons project to the globus pallidus internus and substantia nigra pars reticulata (see Fig. 1).

A Gray-Box Neural Network Model of Parkinson’s Disease Using Gait Signal

In this study, we focused on the gait of Parkinson’s disease (PD) and presented a gray box model for it. We tried to present a model for basal ganglia structure in order to generate stride time interval signal in model output for healthy and PD states. Because of feedback role of dopamine neurotransmitter in basal ganglia, this part is modelled by “Elman Network”, which is a neural network stru...

Untangling Basal Ganglia Network Dynamics and Function: Role of Dopamine Depletion and Inhibition Investigated in a Spiking Network Model

The basal ganglia are a crucial brain system for behavioral selection, and their function is disturbed in Parkinson's disease (PD), where neurons exhibit inappropriate synchronization and oscillations. We present a spiking neural model of basal ganglia including plausible details on synaptic dynamics, connectivity patterns, neuron behavior, and dopamine effects. Recordings of neuronal activity ...

Competition between feedback loops underlies normal and pathological dynamics in the basal ganglia.

Experiments performed in normal animals suggest that the basal ganglia (BG) are crucial in motor program selection. BG are also involved in movement disorders. In particular, BG neuronal activity in parkinsonian animals and patients is more oscillatory and more synchronous than in normal individuals. We propose a new model for the function and dysfunction of the motor part of BG. We hypothesize...

The Role of Inhibition in Generating and Controlling Parkinson’s Disease Oscillations in the Basal Ganglia

Movement disorders in Parkinson's disease (PD) are commonly associated with slow oscillations and increased synchrony of neuronal activity in the basal ganglia. The neural mechanisms underlying this dynamic network dysfunction, however, are only poorly understood. Here, we show that the strength of inhibitory inputs from striatum to globus pallidus external (GPe) is a key parameter controlling ...

A Population of Indirect Pathway Striatal Projection Neurons Is Selectively Entrained to Parkinsonian Beta Oscillations

Classical schemes of basal ganglia organization posit that parkinsonian movement difficulties presenting after striatal dopamine depletion stem from the disproportionate firing rates of spiny projection neurons (SPNs) therein. There remains, however, a pressing need to elucidate striatal SPN firing in the context of the synchronized network oscillations that are abnormally exaggerated in cortic...
